青蒿素及其衍生物(双氢青蒿素,蒿甲醚,青蒿琥脂)是一种具有过氧桥化学结构的倍半萜内酯类化合物,对治疗疟疾有特殊疗效,且具有高效、低毒、成本低、安全等药理特性,使其成为现如今最为广泛应用的一种抗疟药。但是截止目前相关的太赫兹指纹谱数据还存在争议,为该类药物的太赫兹快速、无损鉴定带来困扰。为了进一步完善青蒿素及其衍生物的太赫兹指纹谱数据,利用太赫兹时域光谱(THz-TDS)技术重新测量了青蒿素、双氢青蒿素、青蒿琥酯、蒿甲醚等四种青蒿素类药物在太赫兹波段的吸收特性,分别得到它们在0.2-2.7THz波段的吸收光谱。并对实验结果与现有的指纹谱数据进行了对比分析。结果显示,绝大部分的共振峰与文献[3]中的一致,但峰位和强度略有差异,并且有新的峰位出现。然后,讨论了样品制备和数据处理方法对数据结果的影响,以及分子结构与峰位的关系。并基于此进一步确认了数据的有效性。为进一步研究青蒿素及其衍生物的药性提供参考,并对其无损鉴别、定量定性分析提供了一种快速、有效的检测方法。
Artemisinin and its derivatives:dihydroartemisinin,artemether,artesunate,are a group of sesquiterpene lactone with a 1,2,4-trioxane ring system and possess a rapid action against malaria.Due to its pharmacological features such as efficiency,low toxicity,low cost,safety and so on,they have become one of the most widely used antimalarial drugs.However,there are still some ambiguity in the current terahertz fingerprint spectrum data,which is difficult for the rapid and nondestructive identification of these drugs.In order to further improve the terahertz fingerprint data of artemisinin and its derivatives,here,the terahertz time-domain spectroscopy(THz TDS)was used to measure the THz spectra of artemisinin,dihydro artemisinin,artesunate and artemether,and the characteristic absorption spectra were obtained in the range of 0.2to 2.7THz.Compared with the experimental results and the previous reported data,our experimental data showed that most of the resonance peaks were consistent with the literature[3],but the peak position and intensity were slightly different,and some new peaks were observed.Furthermore,the influence of sample preparation,data processing method on the results and the relationship between molecular structure and peak position were discussed,and the validity of the data was confirmed.This study provides a reference for the further study on the properties of artemisinin and its derivatives,and provides a rapid and effective method for the identification and quantitative analysis of artemisinin and its derivatives.
