目的探讨反义tankyrase 1基因转染对SGC-7901胃癌细胞形态学的影响。方法构建人tankyrase 1基因反义真核表达载体,采用脂质体法将其转染至SGC-7901胃癌细胞中,从光镜、电镜水平观察转染与未转染细胞形态学的变化。结果与亲本细胞相比,光镜下反义tankyrase 1基因转染的SGC-7901细胞胞体增大,胞浆丰富,核浆比例降低,核分裂象减少,核型变规则;透射电镜下核异型性减少,核膜较规则,细胞器丰富,部分细胞胞质内有微腺腔形成。结论反义Tankyrase 1基因转染能部分逆转SGC-7901细胞的恶性表型,有促进其分化的作用。
Objective To study the effects of the transfection of antisense human tankyrase 1 on the morphology of SGC-7901.Method Antisense tankyrase 1 eukaryotic expression on vector pcDNA3.1 was constructed with recombinant DNA technique and transfected into SGC-7901 with liposome DOTAP.Morphological changes of the transfected and non-transfected cells were examined under light and electron microscopy.Results Under light microscopy,transfected cells with antisense gene to tankyrase 1 were larger with abundant cytoplasm and had less nuclei division.The ratio of nuclei to cytoplasm decreased as compared with their parent cells.Under electron microscopy,organelles in these cells were abundant and nuclear membrane was more regular.Conclusion Tankyrase 1 antisense gene can partially reverse malignant phenotypes of SGC-7901 cells and promote its differentiation.
