西太平洋放线菌的分离筛选

放线菌能够产生抗生素、酶及酶抑制剂等,对当代医药工业的发展做出了重大贡献,是最具经济价值和生物技术价值的原核生物。近年来,海洋放线菌及其次生代谢产物的研究也在逐步深入。本文以海洋六号大洋27航次采集的西太平洋样品为基础获得纯培养菌株,结合KOH检测、放线菌特异性引物PCR、BOX-PCR等方法对分离菌株进行初步筛选,然后选取代表性菌株进行16S rDNA测序,共获得了80株多样性较强的海洋放线菌,其中还包括了6株放线菌潜在新属或新种,丰富了西太平洋可培养放线菌的多样性。

英文摘要:

Actinobacteria,the major producer of bioactive microbial secondary metabolites,is the most economically and biotechnologically valuable prokaryote. Recent years have witnessed considerable progress in the investigation of the marine actinobacteria and their secondary metabolites. In the present study,marine actinobacterias were isolated by using samples that collected from the western Pacific.Firstly,actinomycetes strains were obtained by cultured biological method,then the repetitive strains were eliminated according to the morphologic observation,along with the test of KOH solution,BOX-PCR and so on. Finally,the taxonomic status of the representative isolates were preliminarily identified based on 16 s rDNA sequences. By using the methods mentioned above,totals of 80 marine actinomycetes were obtained which includes six potential new genus and new species of actinomycetes. The diversity of the Western Pacific culturable actinomycetes can be enriched.
