目的:观察化痰消瘤方对Lewis肺癌转移的抑制作用,及对E-cad、CD44、CD44v6表达的影响,探讨其作用机制。方法:建立Lewis肺癌小鼠模型,随机分为化痰消瘤方低剂量、中剂量、高剂量组,环磷酰胺组及空白组5组。给药14 d后处死,剥离瘤组织及肺脏,计算各组抑瘤率及抗肺癌转移率。通过逆转录-聚合酶链式(reverse transcripticon-polymerase chain reaction,RT-PCR)、Western blot方法计算化痰消瘤方低剂量、中剂量、高剂量组抑瘤及抗肺癌转移效果最好组与环磷酰胺组及空白组在E-cad、CD44、CD44v6表达上的差异。结果:与空白组比较,化痰消瘤方低剂量组的平均瘤质量及肺转移灶无显著差异(P〉0.05)。化痰消瘤方中剂量、高剂量组及环磷酰胺组平均瘤质量及肺转移灶均低于空白组,且化痰消瘤方抑瘤及抗肺癌转移作用表现出剂量依赖性(P〈0.05)。RT-PCR结果显示:与空白组Lewis肺癌小鼠比较,化痰消瘤方高剂量组及环磷酰胺组的CD44 m RNA、CD44v6 m RNA表达水平显著下调(P〈0.01),E-cad m RNA表达水平显著上调(P〈0.01)。Western blot检测结果显示:与空白组比较,化痰消瘤方高剂量组及环磷酰胺组的CD44、CD44v6的蛋白表达均显著上调(P〈0.01),E-cad的蛋白表达均显著下调(P〈0.01)。与RT-PCR的检测结果表现出相同的趋势。结论:化痰消瘤方具有一定的抑瘤作用及抗肺癌转移作用,且以化痰消瘤方高剂量组疗效显著。其作用机制可能与上调肿瘤细胞E-cad的表达,从而增加肿瘤细胞间的黏附,减弱肿瘤细胞向周围组织侵袭的能力有关;同时还可下调肿瘤细胞CD44、CD44v6的表达,降低肿瘤细胞与基质间的黏附,从而抑制肿瘤细胞的侵袭、转移。
Objective:To study the effect of anti-metastasis and expressions of CD44,CD44v6 and E-cad of Huatan Xiaoliu Decoction in Lewis lung cancer,and to observe the mechanism of restraining tumor growth and tumor metastasis.Methods:The experimental animal Lewis lung carcinoma were randomly divided into low,medium and high dose,CTX and blank groups.After 14days,the experiment was over and the mice were killed.And the tumor was stripped and weighed to calculate the inhibition rate.The weight of lung in mice were counted to calculate the rate of anti-lung metastasis.To study the effects of Huatan Xiaoliu Decoction on tumor metastasis and expressions of CD44,CD44v6 and E-cad in Lewis lung carcinoma by RT-PCR and Western blot.Results:In comparison with blank group,in terms of weights of tumors and pulmonary metastasis focuses,the low-dose group of Hua-tan Xiaoliu Decoction showing no statistical differences(P〈0.05),the medium and high dose groups of Huatan Xiaoliu Decoction,CTX groups showing statistical difference(P〈0.05).The results of RT-PCR indicating that,in comparison with blank group,the expression of CD44 mR NA and CD44v6 mR NA was low(P〈0.01),the expression of E-cad was high(P〈0.01).The expression of CD44,CD44v6 and E-cad were also detected by Western blot.And the results were similar to former.Conclusion:Huatan Xiaoliu Decoction has the function to restrain tumor growth and tumor metastasis in dose-dependent manner.The underlying mechanisms are possibly related to the increasing of the expression of E-cad.It could raise adhesion among tumor cells and weaken the ability of invasion to surrounding tissue by tumor cells.At the same time,the mechanism also relates to the decreasing in expression of CD44 and CD44v6.It could reduce the adhesion between tumor cells and ECM and inhibit the ability of invasion and metastasis of tumor cells.
