目的探讨eIF_4E基因在非小细胞肺癌(Non small cell lung cancer,NSCLC)表达及与化疗敏感性的关系。方法选用外科手术切除NSCLC组织60例以及癌旁正常组织30例作为对照,通过免疫组化检测癌组织以及正常肺组织中elF-4E的表达。NSCLC患者术后接受榄香烯顺铂治疗至少两周以上。比较elF4E在癌组织以及正常肺组织中的表达、eIF_4E阴性表达患者和阳性表达患者的总生存期和化疗效果。结果eIF_4E在NSCLC患者癌组织中的阳性表达率显著高于癌旁组织(P〈0.01);elF-4E阴性表达患者的Os较阳性患者明显延长,阳性表达患者的化疗完全缓解率亦显著低于eIF_4E阴性表达患者(P〈0.01)。结论eIF-4E可能成为NSCLC诊断的标志物,eIF_4E表达水平检测可用于NSCLC患者个体治疗方案的选择。
Objective To understand the expression of elF-4 e gene in non-small cell lung cancer (NSCLC) and its correlation to chemotherapy sensitivity. Method Sample tissues obtained by surgical excision from 60 cases of NSCLC and sample normal tissues adjacent to the carcinoma obtained from 30 cases were chosen for examination. The elF-4E expression in both type of tissues were then subjected to immunohistoehemical analysis. The patients were treated with elemene cisplatin postoperative chemotherapy for a period of more than 2 weeks. The expression of elF-4E, overall survival (OS) and curative effect were analyzed. Findings Positive expression rate of elF-4e in cancerous tissue was found significantly higher than that of the tissues adjacent to carcinoma (P 〈 0.01 ). OS in patients with elF-4e negatively expressed was found significantly longer than that of patients with elF-4e positively expressed, and complete remission rate in elF-4 e negatively expressed patients is significantly higher than that of elF-4e positively expressed patients( P 〈 0.01 ). Conclusion EIF-4e can serve as an indicator of lung cancer. The expression of eIF4E gene is correlated to tumorgenesis, invasion and metastasis of lung cancer. The elF-4e expression level detection can be used for the choice of individual treatment for NSCLC patients.
