目的识别GATA4基因在中国汉族先天性心脏病人群中的变异。方法对348例散发型先天性心脏病病例的GATA4基因进行直接测序筛查,以寻找新的变异。结果在2例先天性心脏病患者中发现2个不同的杂合错义突变(G21V和P407Q),其中G21V为首次报道。GATA4基因在本次研究中的总突变率为0.6%。结论结合之前的研究,本研究更进一步证实GATA4基因在心脏发育及先天性心脏病的发病机制中具有重要作用。
Objective To unfold GATA4 gene variations in Chinese Han population with sporadic congenital heart disease (CHD). Methods GATA4 were screened in 348 individuals with sporadic CHD by direct DNA sequencing. Results Two heterozygous missense mutations (G21 V, P407Q) have been detected in two patients, and G21V is a new mutation, reported for the first time. The overall mutation rate of GATA4 in our study is 0.6%. Conclusion Consistent with previous studies, our study further confirmed that GATA4 plays an important role in normal heart development and pathogenesis of congenital heart disease.
