目的:观察小干扰RNA(siRNA)沉默FBXO22基因对人肾癌细胞株(786-O、ACHN)细胞迁移、侵袭能力的影响。方法分别用siRNA-Ctrl和siRNA-FBXO22小干扰RNA转染人肾癌细胞株786-O和ACHN细胞;Western blot实验检测FBXO22蛋白表达量,CCK-8实验检测FBXO22基因对肾癌细胞增殖的影响,Transwell细胞迁移实验和侵袭实验检测沉默FBXO22基因后对786-O和ACHN肾癌细胞迁移能力和细胞侵袭能力的影响,明胶酶谱实验检测细胞分泌基质金属蛋白酶( MMPs)的能力。结果转染FBXO22 siRNA后肾癌786-O及ACHN细胞的FBXO22表达下调,迁移和侵袭能力提高,分泌MMP-9的能力升高,但对肾癌细胞增殖无影响。结论沉默FBXO22基因后,可以增加MMP-9的分泌,从而激活MMP-9信号通路增强肾癌786-O细胞及ACHN细胞的迁移和侵袭的能力。
Objective To investigate the effects of FBXO 22 silencing by small interfering RNA ( siRNA) on the mi-gration and invasion of human renal carcinoma 786-O cells and ACHN cells .Methods The siRNA-control and siR-NA-FBXO22 fragments were transfected into 786-O and ACHN cells .The level of FBXO22 was detected by Western blotting.The effects of FBXO22 silencing on cell proliferation were examined by CCK 8 assay.The effects of FBXO22 si-lencing on the migration and invasion of 786-O and ACHN cells were investigated by Transwell assay .The levels of ma-trix metalloproteinase ( MMPs) were detected by gelatin zymography .Results FBXO22 siRNA could down -regulated the level of FBXO22 in 786-O and ACHN cells, but stimulate the cells′capacities to migration and invasion and en-hance the secretion of MMP -2, without no influence on renal cancer cells .Conclusion FBXO22 silencing can stimu-late the secretion of MMP-9, which would then activate the migration and invasion of renal carcinoma cells .
