目的探讨Snail1基因启动子区的组蛋白修饰与前列腺癌DU145细胞发生上皮间质转化(epithelial-mesenchymal transition,EMT)现象的关系。方法RT-PCR、Western blot及IF法验证TGF—β1诱导前列腺癌细胞发生EMT过程中相关标记物的变化;利用ChIP试验来筛查Snail1启动子区组蛋白修饰及RNA聚合酶Ⅱ的富集区域。结果诱导前列腺癌发生EMT过程中,仅有DU145细胞EMT相关标记物的变化明显(P〈0.05);DU145细胞中Snail1经TGF-β1诱导后出现明显上调的现象;Snail1基因启动子区域的P4位点存在H3K4ME3及RNA聚合酶Ⅱ的富集。结论TGF-母。通过促使Snail1基因启动子区P4位点的H3K4ME3富集导致Snail1表达上调,促进了前列腺癌DU145细胞EMT现象的发生。
Objective To explore the relationship between histone modification of Snaill promoter and EMT in prostate cancer DU145 cells. Methods RT-PCR, Western blot and IF were used to detect the changes of relevant markers in EMT induced by TGF-β1, and ChIP test was used to screening histone modification and RNA polymerase Ⅱ enrichment area of snaill promoter. Results Markers of EMT changed significantly in DU145 cells treated with TGF-β1 ( P 〈 0. 05 ) ; Expression of Snaill was obviously increased ; P4 site of Snaill promoter existed enrichment areas of H3K4ME3 and RNA polymerase Ⅱ. Conclusion TGF-β1 could up-regulate Snail1 expression through enrichment of H3K4ME3 and RNA polymerase 11 in P4 site of Snaill promoter, promoting EMT in DU145 cells.
