目的:筛选出适宜红蓼总黄酮纯化的大孔树脂,研究影响分离纯化的主要因素,确定最佳工艺参数。方法:采用分光光度法测定红蓼总黄酮含量,通过静态及动态吸附-解吸方法筛选最佳树脂,以总黄酮回收率为主要指标,优化工艺条件。结果:回收率和解吸率最佳为D101大孔树脂,最佳工艺条件为:样品浓度2.213 mg·mL-1、pH=4、用量103.3 mg·g-1;3 BV水洗、4 BV50%乙醇洗脱,总黄酮回收率为56.2%,纯度为72.3%。结论:D101大孔树脂用于红蓼中总黄酮的提取纯化,回收率较高,纯度好。
OBJECTIVE To purify the total flavonoids from Polygonum orientle L with macroporous adsorptive resin ana es- tablish optimum process for separating and purifying total flavonoids. METHODS The optimal techniques of separation and purification of flavonoid were obtained by static adsorption and elution tests, and separation techniques were evaluated by meas- uring the concentration of total flavonoids from Polygonum orientle L with ultraviolet spectrophotometer. RESULTS The type D10I macroporous adsorptive resin was the best for the separation and purification of total flavonoids of Polygonurn orientle L. The optimum process condition was sample amount 103.3 mg· g-1, and sample concentration 2. 213 mg·mL 1 at pH4; and elu- ting reagent was 500/4 alcohol with 4 BV, and water dution volume was 3 BV. The recovery rate of total flavonoids reached 56. 2%with the purity of 72. 3%. CONCLUSION To utilize the D101 macroporous adsorptive resin for the separation and pur- ification of the total flavonoids Polygonum orientle L is feasible, shows high recovery rate and purity.
