为了挖掘和利用华山新麦草(Psathyrostachys huashanicaKeng)的优良基因,应用PCR技术对华山新麦草低分子量谷蛋白亚基(LMW-GS)基因进行了分子克隆和序列分析。结果表明,华山新麦草LMW-GS基因(HS-LMW-1)长度为866 bp,通过提交NCBI进行同源性比较,HS-LMW-1与小麦属LMW-GS基因序列的同源性在83%以上,其中与Thinopyrum intermedium的LMW-GS基因序列(GenBank登录号:AY214454)相似性达92%,进一步将其翻译为氨基酸序列,分析结果显示它具有小麦属LMW-GS序列的典型特征,具有完整的编码区,能够编码274个氨基酸的成熟蛋白,与小麦属Glu-B3和Glu-D3位点编码的LMW-GS基因有很高的一致性,表明HS-LMW-1基因可能由华山新麦草的Glu-Ns3位点编码。
Psathyrostachys huashanica Keng.is an endemic species in China and has been listed as an endangered and imperatively protected wild species.In order to find and utilize the advantageous genes of P.huashanica,LMW glutenin subunit(LMW-GS)gene from P.huashanica was cloned by PCR technique and analyzed.The results showed that the length of the LMW glutenin subunit gene of P.huashanica(HS-LMW-1)was 866 bp,BLAST search showed that HS-LMW-1 was over 83% homologies in nucleotide sequence to LMW-GS genes from Triticum,especially 92% homologies to LMW-GS genes from Thinopyrum intermedium,and this proved HS-LMW-1 was the LMW glutenin subunit gene of P.huashanica.The amino acid sequence deduced from HS-LMW-1 has the typical character of LMW-GS from Triticum,and has a comp1ete coding region encoded a mature protein of 274 amino acid residues.It was found that it was highly similar to those of Glu-B3 and Glu-D3 locus in Triticum aestivum,this suggested that HS-LMW-1 might be Glu-Ns3 locus of P.huashanica.
