目的观察补肾代表中药淫羊藿、补骨脂、女贞子、何首乌有效成分对大鼠骨髓间充质干细胞(BMSCs)增殖及向成骨细胞定向分化的影响。方法将65只SPF级SD雌性大鼠随机分为正常对照组、阳性转化液对照组、补肾中药配伍组(补肾中药1组、2组、3组、4组、5组、6组、7组、8组、9组)、非补肾中药对照组(黄芪甲苷组、川芎嗪组)。通过血清药理学方法,对实验大鼠连续灌胃给药3d后,取材获得含药血清,使用各组大鼠含药血清培养骨髓间充质干细胞6d、12d、18d;用茜素红染色对钙化结节进行成骨细胞鉴定;应用MTF法检测骨髓问充质干细胞的增殖率,酶联免疫吸附剂测定(ELISA)法检测碱性磷酸酶(ALP)活性,用实时定量PCR法检测核心结合蛋白因子2(RUNX2)、成骨细胞特异性转录因子Osterix(OSX)的基因表达。结果补肾中药有效成分不同配伍含药血清作用于体外培养骨髓问充质干细胞72h,可以促进骨髓间充质干细胞的增殖。ALP在12d达到高峰值。Real—TimePCR结果显示补肾中药有效成分能上调RUNX2、OSX的基因表达。结论补肾中药有效成分配伍能提高骨髓间充质干细胞增殖率,促进骨髓间充质干细胞分化为成骨细胞,其机制与补肾中药有效成分配伍能上调RUNX2、OSX的基因表达相关。
Objective To observe the impacts on BMSCs proliferation and osteoblast directional dif- ferentiation by the effective components of Chinese herbs for kidney tonification such as yinyanghuo( Epimedi- um Herb),buguzhi( Fructus Psoraleae),nuzhenzi( Fructus Ligustri Lucidi)and heshouwu( Polygonum Multi- fiorum). Methods Sixty - five SPF - grade SD female rats were randomized into a normal control group, a positive transfer solution control group, kidney tonification herbal combination groups ( No. 1 group, No. 2 group,No. 3 group,No. 4 group,No. 5 group,No. 6 group,No. 7 group,No. 8 group and No. 9 group) ,and the other herbal control groups( astragaloside group,ligustrazine group). Serum pharmacology method was adopt- ed. After gastric infusion of medication continuously for 3 days in the experimental rats, the drug - contained serum was collected. BMSCs were cultured with the drug -contained serum in each group for 6 d, 12 d and 18 d separately. Alizarin red staining was used for osteoblast identification of calcified nodules. MT'F assay was used to determine the proliferation rate of BMSCs. ELISA was used to determine ALP activity. Real - time PCR method was used to determine the gene expressions of runt -related transcription factor -2 (RUNX2)and osteo- blast specific transcription factor Osterix(OSX). Results The drug- contained serum,with the different com- bination of the effective components of Chinese herbs for kidney tonification, acted on BMSCs in vitro for 72 h. The results showed that BMSCs proliferation was promoted. ALP was up to the peak value in 12 d. Real - time RPC results indicated that the effective components of Chinese herbs for kidney tonification up - regulated RUNX2 and OSX gene expressions. Conclusion The combination of the effective components of Chinese herbs for kidney tonification improves BMSCs proliferation and osteoblast differentiation. The mechanism is related to the up - regulation of the gene expressions of RUNX2 and OSX.
