目的:观察玄神活血消肿方对急性软组织损伤大鼠白介素-6(interleukin-6,IL-6)、白介素-1β(interleukin-1β,IL-1β)和血液流变学指标的影响。方法:将60只SD大鼠随机分为阳性对照组、空白对照组、模型组、玄神活血消肿方低剂量组、玄神活血消肿方中剂量组、玄神活血消肿方高剂量组,每组各10只。除空白对照组,其他各组大鼠均采用局部打击法造成急性软组织损伤模型,造模成功后第2天开始给药。玄神活血消肿方低剂量组、中剂量组、高剂量组分别给予玄神活血消肿液0.8g·mL-1,1.6g·mL-1,2.4g·mL-1,每次2mL,灌胃,每日1次。阳性对照组大鼠给予阿司匹林治疗,将阿司匹林肠溶缓释片制成37.5g·L-1的溶液,每次灌胃2mL,每日1次。空白对照组、模型组大鼠分别给予生理盐水。于第7天测量大鼠痛闽,并取损伤部位软组织,匀浆后测定IL-6和IL-1p水平,同时在第7天麻醉大鼠后心脏取血,测定血液流变学情况并进行统计学分析。结果:第7天给药后30rain,玄神活血消肿方低剂量组、中剂量组、高剂量组与模型组比较,痛阈值均显著延长,其中玄神活血消肿方高剂量组痛阈值明显高于模型组,差异具有统计学意义(P〈0.05)。给药后各组大鼠IL-6、IL-1β水平较模型组显著降低,差异具有统计学意义(P〈0.05);玄神活血消肿方各剂量组血浆黏度比、血小板黏附率、血栓长度与模型组比较明显降低,差异具有统计学意义(P〈0.05)。结论:玄神活血消肿方辨治大鼠急性软组织损伤模型疗效显著,其病理机制与抑制炎性细胞因子和改善血液流变学有关。
Objective :To observe the influence of Xuanshen Huoxue Xiaozhong Decoction (XSHXXZD) on interleukin-6 ( IL-6 ) and interleukin-1β (IL-1β) and blood theology indexes of rats with acute soft tissue injury. Methods :Sixty SD rats were randomly divided into the positive control group,the blank control group,the model group,XSHXXZD low-dose group,XSHXXZD middle- dose group,and XSHXXZD high-dose group,with 10 cases in each group. Except the blank control group,the ones in the other groups were made acute soft tissue injury models by using local attack method. Starting from the second day after successful estab- lishment of models, they were given medicine for treatment. The ones in XSHXXZD low-dose group, XSHXXZD middle-dose group,and XSHXXZD high-dose group were given 0. 8 g crude drug · mL-1 of XSHXXZD, 1.6 g crude drug · mL-1 of XSHXXZD,2.4 g crude drug· mL-1 of XSHXXZD respectively for gavage,2 mL each time, and once daily. The ones in the posi- tive control group were given aspirin. Aspirin Enteric-coated Sustained Release Tablets were made into 37.5 mg · mL-1 solution for garage ,2 mL each time, and once daily. The ones in the blank control group and the model group were given normal saline re- spectively. On the seventh day the rats' pain threshold were measured;soft tissues of the injuries were taken, then IL-6 and IL-1β levels were determined after homogenate. At the same time, after the rats were anesthetized rats, heart blood was drawn in order to determine blood theology indexes which would be statistically analyzed. Results:Thirty minutes later after taking drugs on the 7th day, compared with the model group, the pain threshold values of XSHXXZD low-dose group, XSHXXZD middle-dose group, and XSHXXZD high-dose group were significantly prolonged;XSHXXZD high-dose group was significantly higher than the model group, the difference between them was statistically significant ( P 〈 0.05 ).
