为探寻mi RNA在大白菜雄蕊育性调控中的作用,以大白菜雄性不育"两用系"AB03的可育株与不育株花蕾为试材,构建small RNA测序文库并进行高通量测序,最终在两样本中分别检测到132和144个已知mi RNA,其中有19个mi RNA在不育株与可育株花蕾中差异表达。在这19个差异表达的mi RNA中,14个在不育花蕾中上调表达,5个在不育花蕾中下调表达。这些差异表达的mi RNA的靶基因部分与花、雄蕊、花粉的发育相关,例如AP2、果胶甲酯酶抑制剂家族蛋白等,本研究探讨了可能参与花粉发育的mi RNA及其与靶基因的关系。
In order to explore the role of mi RNAs in the regulation of fertility of stamens, this study used the fertile and sterile buds of an AB line of Chinese cabbage, ’AB03’, as the test material, and constructed small RNA libraries. Using high-throughput sequencing, we obtained 132 and 144 known mi RNAs in two samples respectively, and identified 19 differentially expressed mi RNAs. In these 19 differentially expressed mi RNAs, 14 had high expression level in the sterile buds, only 5 were expressed highly in fertile buds. These differentially expressed mi RNAs target genes, which are related to the development of flower, stamens and pollen, such as AP2, pectin methylesterase and so on. The study found that mi RNAs may be involved in pollen development and provided the relationship between mi RNAs and its target genes.
