目的研究小檗碱对糖尿病大鼠心肌缺血再灌注损伤的保护作用及其相关机制。方法实验采用腹腔注射链脲佐菌素65mg/kg复制糖尿病大鼠模型,实验共分5组,糖尿病大鼠假手术组;糖尿病大鼠模型组;小檗碱5、10、20mg/kg3个剂量组。用BL-420生物采集系统记录各组大鼠心率、左宣收缩压、左室舒张末压、左室压力变化速率;用试剂盒测量血清中乳酸脱氢酶和肌酸激酶同功酶活性以及心肌组织中超氧化物歧化酶活性和丙二醛含量。结果小檗碱可升高糖尿病大鼠缺血再灌注后心室收缩压和左室压力变化速率。降低左室舒张末压,降低血清乳酸脱氢酶和肌酸激酶同功酶活性,降低心肌组织丙二醛含量和升高超氧化物歧化酶活性。结论小檗碱对糖尿病大鼠缺血再灌注损伤具有保护作用。
Objective To investigate the protective effects of Berberine on myocardium ischemia/reperfusion in diabetic rats. Methods Diabetic rats was induced by intraperitoneal injection of streptozotocin(STZ) 65mg/kg. All diabetic rats were randomly divided into five groups, sham group; model group ; three Berberine treatment groups (5,10,20mg/kg). The levels of heart rate (HR) , left ventricular systolic pressure (LVSP), Left ventricular end diastolic pressure (LVEDP) and ± dp/dtmax were recorded by BL - 420 system. The levels of lactate dehydrogenase (LDH) and MB isoenzyme of creatine kinase ( CK - MB ) in serum and superoxide dismutase ( SOD ) and malondialdehyde ( MDA ) in myocardial tissues were detected using kits. Results Berberine increased the levels of LVSP, ± dp/ dtmax, and decreased the level of LVEDP. Berberine decreased the content of LDH and CK - MB in serum. Berberine decreased the content of MDA, and increased the activity of SOD in myocardial tissues. Conclusion Berberine can protect diabetic rats cardiomyocytes from ischemia/reperfusion injury.
