目的 探讨不同级别神经胶质瘤中支链氨基酸氨基转移酶1(BCAT1)的表达水平及其与肿瘤增殖的关系.方法 收集首都医科大学附属北京天坛医院神经外科2012至2013年手术切除的54例神经胶质瘤标本,免疫组化检测54例神经胶质瘤中BCAT1和Ki-67的表达水平;实时荧光定量PCR检测BCAT1基因的表达程度.结果 (1)BCAT1表达水平随着病理分级的增高而增多(P<0.05);(2)其表达程度与Ki-67表达呈正相关(r=0.825,P<0.01);(3)随着胶质瘤恶性程度的增高,BCAT1 mRNA的表达增高,WHO Ⅰ级(0.5587±0.2038)和Ⅱ级(1.3499±0.3964)之间的表达差异具有统计学意义(P<0.01),WHOⅡ级和Ⅲ级之间的表达差异具有统计学意义(P<0.01),WHOⅢ级(2.0004±0.4025)和Ⅳ级(3.0656±0.4955)之间的表达差异具有统计学意义(P<0.01).结论 BCAT1与神经胶质瘤恶性程度呈正相关,恶性程度越高其BCAT1表达水平越高.
Objective To investigate the expression levels of branched-chain amino acid transaminase 1 (BCAT1) and its relationship with glioma proliferation in different levels of glioma.Methods The glioma specimens of 54 patients resected at Department of Neurosurgery,Beijing Tiantan Hospital,Capital Medical University from 2012 to 2013 were collected.Immunohistochemistry assay was used to detect the expression levels of BCAT1 and Ki-67 in 54 patients with glioma.Real-time fluorescence quantitative PCR was used to detect the expression degree of BCAT1 gene.Results (1) The expression levels of BCAT1 increased with the elevated pathologic grades (P 〈 0.05).(2) Its expression degree is positively correlated with that of Ki-67 (r =0.825,P 〈 0.01).(3) With the increase of the malignant degree of gliomas,the expression of BCAT1 mRNA increased.There was significant difference in the expression levels between the WHO grade Ⅰ (0.5587 ± 0.2038) and grade Ⅱ (1.3499 ± 0.3964) (P 〈 0.01).There was significant difference in the expression levels between the WHO grade Ⅱ and grade Ⅲ (P 〈0.01).There was significant difference in the expression levels between the WHO grade Ⅲ (2.0004 ±0.4025) and grade Ⅳ (3.0656±0.4955) (P〈0.01).Conclusions BCAT1 is positively correlated with the malignant degree of gliomas.The higher the degree of malignancy,the higher the expression level of BCAT1.
