目的探讨miR-185(HomosapiensmiR-185,miR-185)在结肠癌组织中的表达及其临床意义。方法运用原位杂交技术(hybridizationinsitu,ISH)检测miR-185在结肠癌及癌旁组织中的表达情况,分析其表达与结肠癌临床分期及淋巴结转移间的关系。结果ISH检测显示,在52例结肠癌组织中miR-185阳性表达率为13.46%,与癌旁对照组(84.62%)相比,差异有显著性(P〈0.01);miR-185在结肠癌临床Ⅲ/Ⅳ期、Ⅰ/Ⅱ期和癌旁对照组中的光密度值分别为0.12±0.01、0.37±0.04、0.72±0.09,临床Ⅲ/Ⅳ期、Ⅰ/Ⅱ期分别与癌旁对照组比较,差异有显著性(P〈0.05);淋巴结转移的与无淋巴结转移miR-185表达的光密度值分别为0.11±0.02、0.39±0.04,有淋巴结转移的miR-185表达水平低于无淋巴结转移,两者差异有显著性(P〈0.01)。miR-185的表达随着结肠癌临床分期演进而降低。结论nqiR-185与结肠癌的发生、临床进展、转移密切相关,其可能成为结肠癌治疗、预后判断的潜在生物学指标。
Objective To investigate the expression of the miR-185 in tissue of colon carcinoma and its clinical significance. Methods The tissue specimens and paracancerous tissues of 52 cases were collected and compared. Hybridization in situ was nsed to detect the miR-185 expression in those specimens. Results The positive expression rate of the miR-185 was 13. 46% in colon carcinoma which was lower than the control group (84. 62% ) (P 〈 0. 01 ). It suggested that the miR-185 expression was associated with advanced clinical stage and with lymph node metastasis, stage III/IV were higher than I/II tumor( P 〈 0. 05) ,tumors with lymph node metastasis were lower than tumors without lymph node metastasis in colon carcinoma (P 〈 0. 01 ). Conclusions MiR-185 may play an important role in the proliferation,and metastasis of colon carcinoma,and it could be an indieator in the diagnosis and prognosis of colon carcinoma,miR-185 can be used as a predictive biomarker.
