目的筛选促进金线莲生长发育的优良内生真菌。方法在离体培养条件下,将金线莲试管苗与4株分离自兰科植物的内生真菌在5种供试培养基上共培养,根据菌苗共生特点筛选适宜的双重培养基。在此基础上,将17株分离自兰科植物的真菌与金线莲苗共生培养,以鲜重和增高为指标,筛选可促进金线莲生长发育的内生真菌。结果17株真菌中有1株显著促进金线莲苗增高(P〈0.05),3株极显著促进金线莲苗增高(P〈0.01),最高促增高可达对照的2.04倍,其中2株为金线莲有益真菌。结论双重培养基和金线莲有益真菌的筛选为研究内生真菌和金线莲的共生机制及人工栽培金线莲新方法奠定了基础。
OBJECTIVE To select the endophytic fungi promoting growth and development ofAnoectochilus roxburghii . METHODS A. roxburgii seedlings and 4 strains endophytic fungi isolated from the orchids were cultured together on five kinds of medium in vitro culture. Suitable dual culture medium were selected according to the growth of the plantlets and fungi. On the basis of these, 17 strains of fungi isolated from orchids were cultured together with seedlings for one month, respectively. Endophytic fungi promoting growth and development of A. roxburgii were selected, with its fresh weight, plant height as index. RESULTS One strain could promote seedlings growth significantly (P〈0.05), and three strains extremely significantly increased the plant height of A. roxburghii seedlings(P〈0.01), which could reach as high as 2.04 times compared with the control. Two strains of them showed effective influence on A. roxburghii growth. CONCLUSION The selection of dual culture medium and beneficial fungi of A.roxburghii laid a foundation for the further research of the symbiotic mechanism between them and new artificial cultivation methods of A. roxburghii.
