为分析碳源对嗜热β-半乳糖苷酶在组成型重组毕赤酵母中的表达水平,葡萄糖、果糖、丙三醇、乳糖、蔗糖和麦芽糖分别用于配制培养基的碳源.当以单糖和甘油为碳源时,酵母的生长和重组嗜热β-半乳糖苷酶的分泌水平要优于双糖;以果糖为碳源时,重组酵母分泌嗜热β-半乳糖苷酶的水平最高,其次为葡萄糖和丙三醇,蔗糖要高于麦芽糖,乳糖最低.
To analyze the thermostabiliuy of recombinant β- Galactosidase secretory expression level in the constitutive recombinant Pichia pastoris, glucose, fructose- glycerol, lactose, sucrose and maltose were used as carbon sources respectively. The results showed that monosaccharide and glycerol were better than disaccharide in cell growth and secretory expression of recombinant β- Galactosidase. The secretory expression level of thermostability recombinant β- Galactosidase in fructose was best than the other 5 carbon resources, glucose second and followed by glycerol, sucrose better than maltose,lactose was the lowest.
