目的比较鼻咽癌高癌家系不同体质证型癌患者鼻咽癌(nasopharyngealcarcinoma,NPc)组织蛋白质组差异,筛选体质证型相关性差异蛋白质。方法鼻咽癌高癌家系初诊鼻咽癌患者14例,包括气虚体质组7例和湿热体质组7例,取鼻咽原发灶癌组织,应用双向凝胶电泳技术分离标本组织总蛋白后,自动识别差异表达蛋白质点,基质辅助激光解吸电离飞行时间质谱MALDI-TOF-MS分析得到肽质量指纹图谱PMF,生物信息学技术鉴定部分差异蛋白质点及其病理意义。结果建立了鼻咽癌高癌家系癌患者气虚体质组和湿热体质组鼻咽原发灶癌组织的双向凝胶电泳图谱,发现该系气虚体质组鼻咽癌患者和湿热体质组患者之间差异表达大于3倍的蛋白质点10个,并鉴定出了F一肌动蛋白戴帽蛋白亚基(Factincappingprotein)等5个蛋白质。这些差异表达蛋白质的功能涉及遗传与转录、物质代谢、能量的产生、物质转运等现象。结论鼻咽癌高癌家系气虚体质鼻咽癌患者组和湿热体质鼻咽癌患者鼻咽病灶组织存在不同的蛋白质表达谱,已经筛选鉴定的5个差异表达蛋白质,可能为揭示鼻咽癌高癌家系不同体质患者鼻咽癌发病机制研究提供有用信息。
Objective To screen physically associated differentialy expressive proteins of nasopharyngeal focus tissue samples among cases with nasopharyngeal carcinoma (NPC) in different physical pattern from NPC highly occurring family in a comparative way.Methods To screen physically associated differentialy expressive proteins of nasopharyngeal focus tissue samples among cases with nasopharyngeai carcinoma (NPC) in different physical pattern from NPC highly occurring family in a comparative way. Results Established was the database of physical pattern associated 2 DE maps through comparative investigate on nasopharyngeal focus tissue samples from NPC cases occurring in NPC highly occurring families in different physical patterns i.e. Qi deficiency physical pattern and dampness heat physical pattern. Ten differentially expressed proteins were found in these samples with their expressive activities higher than 3 times, when compared between these two groups of cases in different physical patterns. Then, 5 proteins were identified, one of them being F actin capping protein. These proteins were involved in these biological activitiesw such as heredity and transcription, substance metabolism, energy production, materiai transfer, etc. Conclusion It should be true that physically associated differentially expressed proteins are present in nasopharyngeal focus tissue of NPC cases in Qi dificient physical pattern and dampness-heat physical pattern, occurring in NPC highly occurring families. On the basis of bioinformatic analysis, identified 5 proteinsmay provide useful information for further research on the pathogenesis of physical pattern related NPC for cases occurred in NPC highly occurring families.
