本试验旨在研究早期补喂瘤胃液对羔羊血浆免疫球蛋白、细胞因子含量和小肠黏膜免疫相关细胞数量的影响。选取12只特克赛尔(Texel)初生羔羊,随机分成2组,每组6只。试验组羔羊出生后连续补喂瘤胃液14 d,每天1次,对照组羔羊补喂等量生理盐水。羔羊7、14、21和28日龄时,颈静脉采血并分离血浆;28日龄时,屠宰收集肠道内容物。测定血浆中免疫球蛋白[免疫球蛋白A(Ig A)和免疫球蛋白G(Ig G)]、细胞因子[白细胞介素-1β(IL-1β)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)],肠道内容物中Ig G和免疫球蛋白Fc片段(Fc)含量。结果表明:试验组21日龄血浆中Ig A和Ig G含量显著或极显著低于对照组(P〈0.05或P〈0.01),其他日龄组间差异不显著(P〉0.05);试验组血浆中IL-1β和IL-6含量较对照组高,但差异不显著(P〉0.05);试验组血浆TNF-α含量在14日龄时显著高于对照组(P〈0.05),在其他日龄差异不显著(P〉0.05);试验组羔羊血浆IFN-γ含量低于对照组,但差异不显著(P〉0.05);给新生羔羊补喂瘤胃液对28日龄羔羊肠道内容物中Ig G和Fc含量无显著影响(P〉0.05);试验组肠道上皮内淋巴细胞数量较对照组增加,尤其在回肠,与对照组差异显著(P〈0.05);肠道肥大细胞数量各肠段组间无显著差异(P〉0.05)。上述结果表明,早期补喂瘤胃液对羔羊体液免疫无显著影响,可在一定程度上增强由免疫细胞介导的免疫功能;早期补喂瘤胃液对羔羊小肠转运母源免疫球蛋白无显著影响;早期补喂瘤胃液能刺激羔羊小肠黏膜免疫系统的发育,使免疫屏障功能尽早完善。
This experiment aimed to study the effects of early administration of ruminal fluid on plasma immunoglobins and cytokines,and mucosal immunity-associated cells in small intestine in lambs.Twelve Texel newborn lambs were randomly allotted to two groups with 6 lambs per group.Ruminal fluid was orally administered once daily over a period of 14 days in trial group,and physiological saline was administered in control group.Blood was collected from jugular vein to separate plasma at 7,14,21 and 28 days of age,respectively;lambs were slaughtered to obtain small intestinal digesta at 28 days of age.Immunoglobins[immunoglobin A(IgA) and immunoglobin G(IgG)]and cytokines[interleukin-1β(IL-1β),interleukin-6(IL-6),tumor necrosis factor α(TNF-α) and interferon γ(IFN-γ)]contents in plasma,and IgG and immunoglobin Fc fragment(Fc) contents in intestinal digesta were determined.The results showed as follows:at 21 days of age,the contents of IgA and IgG in plasma in trial group was significantly lower than those in control group(P〈0.05 or P〈0.01),but there were no significant differences at the other days of ages(P〉0.05);the contents of IL-1β and IL-6 in plasma in trial group were higher than those in control group,but the differences were not significant(P〉0.05);plasma TNF-α content in trial group was significantly higher than that in control group at 14 days of age(P〈0.05),but there were no significant differences at the other days of ages(P〉0.05);plasma IFN-γ content in trial group was higher than that in control group,but the difference was not significant(P〉0.05);there was no significant difference of IgG and Fc contents in intestinal digesta at 28 days of age(P〉0.05);the administration of ruminal fluid increased intestinal intraepithelial lymphocyte count,especially in ileum,trial group was significantly different from control group(P〈0.05);mast cell count at different intestinal segments was not significantly different between groups?
