目的研究佐剂关节炎(AA)大鼠心肺功能变化与NF-kBp65/TNF-α、TGF-β1/Smad通路的关系。方法将30只大鼠随机均分为正常对照(NC)组和模型对照(MC)组,向MC组大鼠右后足跖皮内注射弗氏完全佐剂0.1ml致炎,复制成AA模型。致炎19d后,观察大鼠足跖肿胀度、关节炎指数(AI),采用超声诊断仪检测大鼠心功能、小动物肺功能仪检测肺功能变化,免疫印迹法检测大鼠心肺NF-kBp65/TNF-α、TGF-β1、Smad2、Smad4、Smad7蛋白表达。结果与正常组比较,AA大鼠足趾肿胀,AI升高,心肺功能参数降低,心肺组织NF-kBp65/TNF-α、TGF-β1、Smad2、Smad4蛋白表达升高,Smad7降低(P〈0.01或P〈0.05);相关性分析显示,AA大鼠心肺功能参数与NF-kBp65/TNF-α、TGF-β1,smad通路存在关联。结论AA大鼠心肺功能损伤可能与NF-kBp65/TNF-α、TGF-β1/Smad通路过度激活有关。
NF-kBp65/TNF-α、TGF-β1/Smad pathway imbalance plays an important role in rheumatoid arthritis. To evaluate the correlation of cardiopulmonary function change with NF-kBp65/TNF-α、TGF-β1/Smad pathway in rats of adjuvant arthritis (AA), total of 30 rats were randomly divided into normal control (NC) and model control (MC) groups in this study. The MC rats were injected with 0.1 ml Freund's complete adjuvant in the right hind limb to induce inflammation, and 19 days later, we observed paw swelling and arthritis index (AI), detected cardiac function using ultrasonic diagnostic equipment, tested pulmonary function using small animal spirometer, detected the expression of NF-kBp65/TNF-α、TGF-β1, Smad2, Smad4, and Smad7 protein by Western blot. We found that paw swelling, AI, NF-kBp65/TNF-α、TGF-β1, Smad2, Smad4 protein levels increased, but cardiopulmonary function parameters and Smad7 protein level decreased in the MC rats (P 〈0.01 or P 〈 0.05). Correlation analysis showed that the cardiopulmonary function parameters were associated with NF-kBp65/TNF-α、TGF-β1/Smad pathway. In conclusion, the cardiopulmonary function impairment may be associated with excessive activation of NF-NF-kBp65/TNF-α、TGF-β1/Smad signaling pathways.
