杜仲皮和叶中黄酮的含量与抗氧化活性比较

目的对杜仲皮和叶中的黄酮进行提取、纯化及性质分析。方法以50%的乙醇试剂作为浸提液。通过超声波辅助提取杜仲皮和叶中的黄酮。然后利用聚酰胺柱层析,分别用15%、30%、55%的乙醇试剂为洗脱液进行梯度洗脱,洗脱液经过浓缩、冻干,所得杜仲皮黄酮的样品分别为S1、S2、S3,杜仲叶黄酮的样品分别为S4、S5、S6。对所有样品进行含量测定和抗氧化活性分析。结果S1、S2、S3、S4、S5、S6黄酮含量分别为25.93%,33.89%,33.89%,24.68%,41.50%,31.80%;S1、s2、S3、S4、S5、s6半抑制率EC50分别为1.948,2.379,3.133,1.819,2.368,3.312mg/mL。结论杜仲皮和杜仲叶中的黄酮含量接近,抗氧化活性也接近,可以叶代皮做药源。

英文摘要:

Objective To study the extraction, purification and antioxidant activities of Flavonoids from Eucommia Leaves and barks. Methods Total Flavonoids from Eucommia Barks and Leaves were extrac- ted with 50% ethanol solution by the ultrasonic method. The extraction was purified by polyamide col- umn chromatography with different elution including 15%, 30% and 55% ethanol solution. After the ro- tary evaporation and vacuum freeze -drying, the powder samples from Eucommia Barks were named Sl , S2 and S3 And the powder:sample from Eucommia Leaves were named S4, S5 andS6, respectively. The contents and antioxidant activities of Flavonoids were detected. Results The contents of Flavonoids in S1 ,S2, S3, S4, S5 and S6 were 25.93%, 33.89%, 33.89%, 24.68%, 41.50% and 31.80% , respec- tively. The EC50 of S1, $2, $3, $4, S5 and S6 were 1. 948, 2. 379, 3. 133, 1. 819, 2. 368 and 3. 312 mg/mL, respectively. Conclusion Eucommia leaves and barks have similar contents and antioxidant ac- tivities of Flavonoids and Eucommia leaves could be substituted for barks as the raw materials of Fla- vonoids.
