目的探讨整合素和金属蛋白酶域33(ADAM33)基因位点多态性与支气管哮喘易感性的关系。方法检索Pubmed、Web of Sience、Ovid、Embase、Medline、万方数据知识服务平台、超星读秀、维普网、中国知网(CNKI)等电子数据库,检索时间均从建库至2015年10月,检索有关ADAM33基因多态性与支气管哮喘易感性关系的病例对照研究,筛选符合纳入标准的文献,进行数据采集、提取,进行ADAM33基因F+1、T1位点多态性与中国汉族人群支气管哮喘之间关系的Meta分析。结果最终纳入8篇文献,共包括1 718例支气管哮喘患者(病例组)和1 622例健康对照者(对照组)。Meta分析结果显示ADAM33基因F+1位点等位基因C与中国汉族人群支气管哮喘风险降低有关(OR=0.855,95%CI=0.743~0.984,P=0.029)。ADAM33基因T1位点等位基因A能降低中国汉族人群支气管哮喘易患风险(OR=0.408,95%CI=0.254~0.658,P=0.000)。在纯合子模型、显性模型及隐性模型中,ADAM33基因T1位点AA基因型会显著降低中国汉族人群哮喘的发病风险[(OR=0.226,95%CI=0.100~0.511,P=0.000),(OR=0.385,95%CI=0.162~0.918,P=0.031),(OR=0.278,95%CI=0.186~0.415,P=0.000)],而在杂合子模型中未发现与哮喘易患性相关。结论 ADAM33基因F+1、T1位点多态性有可能与中国汉族人群支气管哮喘发病风险降低有关。
Objective To explore the correlation of ADAM33(a disintegrin and metalloproteinase domain33)gene polymorphisms with asthma.Methods An electronic search of the PubMed,Web of science,Ovid,Embase,Medline,WanFang,ChaoXing,VIP database,CNKI and other electronic databases was performed from their foundation to October 2015.Retrieved case-control studies related to association of ADAM33 polymorphisms with asthma,and screened the included studies up to standard,collected and extracted data.A meta-analysis was conducted on correlation between the ADAM33F+1and T1 polymorphisms with asthma.Results A total of eight case-control studies were included,which were including1718 asthma patients and 1622 healthy controls.Meta-analysis revealed that ADAM33 gene F+1Callele decreased the risk of susceptibility to asthma in Chinese Han population(OR =0.855,95%CI =0.743-0.984,P =0.029).ADAM33 gene T1Aallele also decreased asthma risk in Chinese Han population(OR=0.408,95%CI =0.254-0.658,P =0.000).ADAM33 gene T1AA genotype significantly decreased asthma risk in Chinese Han population in additive model,dominant model and recessive model[(OR =0.226,95%CI=0.100-0.511,P =0.000),(OR =0.385,95%CI=0.162-0.918,P =0.031),(OR=0.278,95%CI =0.186-0.415,P =0.000)].However,in codominant model,there was no correlation found between bronchial asthma and ADAM33T1 polymorphism.Conclusion ADAM33 gene F+1and T1 polymorphisms may decrease asthma risk in Chinese Han population.
