目的:探讨酸敏感离子通道(acid sensitive ion channels,ASICs)在大鼠静息态和活化态星形胶质细胞亚细胞分布变化及调控。方法:取大鼠静息态和活化态星形胶质细胞,通过应用免疫荧光、全细胞膜片钳和Western Blot等技术,观察ASICs在静息态和活化态星形胶质细胞亚细胞分布变化及可能机制。结果:(1)ASICs在静息态和活化态星形胶质细胞分别主要分布于细胞核或胞质/胞膜。以ASIC1为例进行分析,发现ASIC1胞核荧光密度在静息态和活化态星形胶质细胞分别为(54.32±1.29)(n=36)和(22.27±0.96)(n=34),而ASIC1胞质/胞膜荧光密度在这两种星形胶质细胞分别为(6.14±0.39)(n=38)和(37.45±1.31)(n=36),这种分布差异在功能学上表现为ASIC1电流的增加,即ASIC电流由静息态的10%增加到活化态的30%;(2)静息态和活化态星形胶质细胞之间ASICs蛋白表达总量差异无统计学意义;(3)活化态星形胶质细胞炎症指示蛋白诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)表达较静息态星形胶质细胞显著增多;用小胶质细胞原性炎症因子白细胞介素1β(interleukin 1β,IL-1β)刺激静息态星形胶质细胞,可使静息态星形胶质细胞ASIC1从胞核转向胞质/胞膜。结论:ASICs在静息态星形胶质细胞主要分布于胞核,而在活化态星形胶质细胞主要分布于胞质/胞膜;IL-1β可能调控星形胶质细胞ASICs由胞核转移至胞质/胞膜。
Objective: To explore the sub-cellular distribution and its regulation of acid sensingion channels(ASICs) in resting and reactive rat astrocytes.Methods: To observe sub-cellular distribution of ASICs and the possible regulated mechanisms by using the techniques of whole cell patch clamp,calcium imaging,Western Blot,and immunofluorescence in resting and reactive rat astrocytes.Results:(1)ASICs are primarily distributed in nucleus or cytoplasm/membrane of resting and reactive astrocytes respectively.Take ASIC1 as an example for analysis,we find that the fluorescence density of ASIC1 in nucleus of resting or reactive astrocytes is(54.32±1.29)(n=36) and(22.27±0.96)(n=34) respectively,while the fluorescence density of ASIC1 in the cytoplasm/membrane of the two kind of cells is(6.14±0.39)(n=38) and(37.45±1.31)(n=36) respectively.This discrepancy of sub-cellular distribution is functionally confirmed by the increase of the current ratio of ASIC1,which is increased from 10% in resting astrocytes to 30% in reactive astrocytes.(2)The total expression of ASICs protein takes no significant variations between resting and reactive astrocytes.(3)The expression of iNOS,a marker for inflammation in reactive astrocytes,is significantly increased.ASIC1 can be transferred from nucleus to cytoplasm by incubating IL-1β,a microglia-derived inflammatory factor,in resting astrocytes.Conclusion: ASICs are mainly distributed in nucleus in resting astrocytes,while primarily located in cytoplasm/membrane in reactive astrocytes.IL-1β might prompt the trafficking of ASICs from the nucleus to the cytoplasm/membrane in astrocytes.
