目的观察五子衍宗丸对肾精亏虚大鼠支持细胞功能的影响。方法 SD大鼠分为正常组、雷公藤多甙组和五子衍宗丸高、中、低剂量组。采用雷公藤多甙每日灌胃制备肾精亏虚大鼠模型,五子衍宗丸灌胃后,分别采用酶免法、HE染色和原位末端标记(TUNEL)测定血清抑制素(INH)B含量、生精细胞Johnson积分和生精细胞凋亡率(AR)。五子衍宗丸含药血清与TM4支持细胞共孵育后,比色法测定细胞色素C氧化酶(COX)活性。结果肾精亏虚大鼠INHB含量和Johnson积分均显著降低,AR显著升高(P〈0.05)。而五子衍宗丸高、中剂量组干预后INHB含量、Johnson积分显著增高、AR显著降低(P〈0.05),高剂量组COX活性显著提高(P〈0.05)。结论五子衍宗丸改善生精细胞功能与其改善支持细胞功能有关。
Objective To observe the influence of Wuziyanzong Pills on the function of Sertoli cells in rats with insufficiency of kidney essence. Methods SD rats were divided into the normal group,glycosides of tripterygium wilfordii group ( GTW group) ,and high-dose,mid-dose and low-dose Wuziyanzong Pills groups ( high-dose group,mid-dose group and low-dose group) . The rat model of insufficiency of kidney essence was established by given GTW intragastrically every day. After Wuziyanzong Pills was given to relevant groups intragastrically,serum inhibin B ( INHB) ,and Johnson scores and apoptosis rate ( AR) of spermatogenic cells were detected by applying ELISA,HE staining and TUNEL. The medical serum of Wuziyanzong Pills and TM4 Sertoli cells were cultured together,and then the activity of cytochrome C oxidase ( COX) was detected by using colorimetry. Results The content of INHB and Johnson scores decreased significantly and AR increased significantly in rats with insufficiency of kidney essence ( P 0. 05) ,while content of INHB and Johnson scores increased significantly and AR decreased significantly in the high-dose group and mid-dose group ( P 0. 05) . The activity of COX increased significantly in the high-dose group ( P 0. 05) . Conclusion The effect of Wuziyanzong Pills to improve the function of spermatogenic cells is related to that it can improve the function of Sertoli cells.
