目的:探讨CisoDGR多肽对乳腺癌MFC-7细胞增殖和凋亡的影响及可能机制。方法:采用MTT法检测CisoDGR多肽对乳腺癌MCF-7细胞的抑制率;流式细胞术检测CisoDGR多肽对MCF-7细胞凋亡的影响;Westernblot法检测Caspase-3、Bcl-2蛋白的表达。结果:CisoDGR多肽能明显抑制乳腺癌MCF-7细胞的增殖,诱导细胞凋亡,并存在一定的时-效及量-效关系。其对凋亡的诱导作用可能与Caspase-3蛋白表达增加及Bcl-2蛋白表达下降有关。结论:CisoDGR多肽具有抑制MCF-7细胞增殖,诱导MCF-7细胞凋亡的作用,其作用机制可能与Caspase-3、Bcl-2蛋白的表达变化有关。
Objective:To investigate the effect of CisoDGR polypeptides on proliferation and apoptosis of human breast cancer MCF -7 cells as well as the relevant mechanism. Methods:MCF -7 cells were tested for proliferation activity by MTT assay, and analyzed for apoptosis rate and cell cycle by flow cytometry. The expressions of Caspase - 3 and Bcl- 2 were detected by Western blot. Results: CisoDGR polypeptides inhibited the cell proliferation and induced apoptosis in MCF - 7 cells in time - and dose - dependent way. The expression level of Bcl - 2 in MCF - 7 cells after treatment with CisoDGR polypeptides decreased significantly, while the level of Caspase - 3 was raised. Conclusion:CisoDGR inhibited the proliferation and promoted the apoptosis of MCF -7 cells by a mechanism which might be related to the changes of expressions of Caspase -3 and Bcl -2.
