目的:筛选与优化银翘散提取工艺,为银翘散复方制剂谱效关系研究奠定基础。方法:采用正交实验筛选银翘散最佳提取条件,分别以水、30%、50%、70%及95%乙醇回流提取银翘散,以高效液相色谱法测定银翘散指纹图谱,以连翘苷、绿原酸、牛蒡苷、橙皮苷等物质的含量作为工艺评价指标,筛选与优化银翘散提取工艺。结果:提取溶剂量及提取次数是影响出银翘散膏量重要因素,其次为提取时间;高效液相色谱法测定不同银翘散提取物均含有27个峰所属化合物。结论:方法简便,重现性好,用于银翘散提取物的鉴别,具有良好的专属性;银翘散各成分之间色谱峰形良好,基本达基线分离,各提取方法的银翘散提取物的某些化学成分及含量有差异,可作为银翘散抗病毒谱效关系研究基础。
Objectlve:To screen and optimize the extraction technology of Lonicerae and Forsythiae Powder and establish the basis for the chromatogram-effectiveness relationship research of the compound preparation, Lonicerae and Forsythiae Powder. Method: To screen the optimized extract conditions of Lonicerae and Forsythiae Powder using the orthogonal test, the compound prepration was refluxed with water,30%,50%,70% and 95% ethanol, respectively, to extract the main compounds of Lonicerae and Forsythiae Powder, in which the four compounds, forsythin, ehlorogenic acid, aretiin and hesperidin, were determined by the high performance liquid chromatography (HPLC) to evaluate the extract technology of Lonicerae and Forsythiae Powder. Results: The quantity of extraction solvent and extract times were the most important factors affecting the extraction number of Lonicerae and Forsythiae Powder, and the next was the extract time. HPLC determines different extract of Lonicerae and Forsythiae Powder, and all the extracts have 27 compounds with good chromatographic peak shapes, the separation of which offer the baseline resolution with adjacent peaks. Some compunds in the each extracts of Lonicerae and Forsythiae Powder were different. Conclustion: This could be used as research basis for the antivirus chromatogram-effectiveness relationship of Lonicerae and Forsythiae Powder.
