目的为探索新基因ndrg2在胎儿小脑组织中表达分布的特点,进一步研究ndrg2功能提供实验依据。方法收集引产4、5个月胎脑组织标本,分别进行组织石蜡切片和总RNA提取,应用免疫组化方法和逆转录酶-多聚酶链反应(RT—PCR)技术检测ndrg2在蛋白质及mRNA表达水平变化规律。结果免疫组化结果表明,在上述胎儿小脑脑区神经元胞浆均有ndrg2蛋白不同程度的表达。RT-PCR结果显示,ndrg2 mRNA在正常胎小脑均有表达,5个月胎儿小脑表达高于4个月胎儿小脑。结论随胎龄增加,ndrg2表达水平增高。
Objective To explore the expression features of ndrg2 in human embryonic cerebellum and provide some clues for further study the function of ndrg2, Methods Human embryonic brain tissues of induceproduction in 4 th and 5 th months were collected. Half of these tissues were embed with paraffin, and the rest half of these tissues were for total RNA extraction. Immunohistochemistry staining and reverse transcriptase polymerase chain reaction (RT-PCR) were used to examine the expression of ndrg2 in protein and mRNA. Results Immunohistochemistry result showed that ndrg2 protein was differently expressed in the neuronal endochylemas of normal 4 th and 5 th months human embryonic cerebellum tissues. RT-PCR result indicated that ndrg2 mRNA was found to be expressed in these normal 4 th and 5 th months human embryonic cerebellum tissues. The expression of ndrg2 in 5 th months embryonic cerebellum was higher than that in 4 th months embryonic cerebellum. Conclusion With the development of human embryonic brain, the expression of ndrg,2 in embryonic cerebellum functional areas increased.
