目的 探讨辛伐他汀对胰腺癌BxPC-3细胞株Shh相关蛋白的影响。方法 胰腺癌BxPC-3细胞株随机分成对照组,5-氟尿嘧啶(5-FU)组(50μmol/L),辛伐他汀高、中、低剂量组(20,10,5μmol/L)。采用台盼蓝染色法和MTT法检测胰腺癌BxPC-3细胞株的生长曲线和增殖抑制率;采用细胞划痕法检测胰腺癌BxPC-3细胞株运动能力;采用免疫组织化学、RT-PCR法检测胰腺癌BxPC-3细胞株Shh、Ptch1、Smo、Gli1水平。结果与对照组比较,5-FU组,辛伐他汀高、中剂量组胰腺癌BxPC-3细胞OD值、运动能力和Shh、Ptch1、Smo、Gli1水平明显降低(P〈0.05)。结论 辛伐他汀通过抑制BxPC-3细胞株Shh、Ptch1、Smo、Gli1水平在胰腺癌治疗中发挥积极作用。
Objective To study the effects of simvastatin on Shh associated protein of BxPC-3 cell line in the pancreatic cancer. Methods BxPC-3 cell line of pancreatic cancer were randomly divided into control, 5-Fluorouracil (5-FU,50 μmol/L), simvastatin high- (20 μmol/L) ,middle-( 10μmol/L), low-dose (5 μmol/L) groups. The trypan blue staining and MTY were used to determine growth curve of BxPC-3 cell line and OD values of pancreatic cancer. The cell scratch was used to determine the exercise capacity of BxPC-3 cell line of pancreatic cancer. Immunohistochemistry (IHC) and real-time PCR (RT-PCR) was applied for detecting Shh, Ptchl, Smo, Glil levels in Bx- PC-3 cell line of pancreatic cancer. Results Compared with the control group, OD values of BxPC-3 cell line of pancreatic cancer, exercise capacity,Shh, Ptchl, Smo, Gill level were significantly decreased in 5-FU, simvastatin high-dose and middle-dose groups( all P〈0.05). Conclusions Simvastatin plays a certain role in the treatment of BxPC-3 cell line of pancreatic cancer by inhibiting Shh, Ptchl, Smo, Glil.
