目的考察灯盏细辛有效组分的的钙拮抗活性。方法采用^45Ca跨膜内流测量技术与兔离体肠肌实验,观察了灯盏细辛中水提物和总黄酮的钙拈抗活性。结果^45Ca跨膜内流测量实验中,与空白对照组比较灯盏细辛总黄酮(0.25、1.0g·L^-1)均有明显的抑制钙离子内流作用(P〈0.05,P〈0.01),水提物浓度为1.0g·L^-1时无钙拮抗作用反而促进钙离子内流。兔离体肠肌实验中,与空白对照组比较水提物组(1.0g·L^-1)的频率,总黄酮组(1、0.5g·L^-1)的频率及张力均有显著抑制作用(P〈0.05)。结论灯盏细辛总黄酮有明显的钙拈抗作用,而其水提物的钙拮抗作用还有待进一步研究证实。
Objective To investigate the Ca^2+ antagonistic effect of active components from Erigeron breviscapus. Methods By 4SCa transmembrane influx method, observed the effect on voltage - dependent Ca^2+ channel (VDC) in rat aorta about aqueous extract and the flavonoids.Moreover, by duodenum of rabbits in vitro, observed the effect about aqueous extract and the flavonoids on the shrink of intestines, which was excited by Ca^2+ .Results Compared with blank control group, the flavonoids (0.25, 1g·L^-1) showed a significant inhibition of Ca^2+ influx ( P 〈 0.05, P 〈 0.01 ), but aqueous extract ( 1 g·L^-1 ) promoted Ca2 + influx ( P 〈 0.01) . Moreover, aqueous extract (1g· L^-1) could reduce the frequency of intestines shrink only, the flavonoids (0.5, 1g· L^-1 ) could reduce both the frequency and tension ( P 〈 0.05) . Conclusion The flavonoids had definite Ca^2+ antagonism, there would need more experiments to verify whether the aqueous has Ca^2+ antagonism.
