目的:痛情绪是疼痛体验的一个重要部分。条件性位置回避(conditioned place avoidance,CPA)装置可用来测量痛情绪,本研究通过操纵影响CPA形成的变量来探究哪些因素会影响痛情绪的形成。方法:采用高渗盐水足底注射建立大鼠条件位置回避(CPA)模型。结果:足底高渗盐水注射引起大鼠自发痛反应,5 min达高峰,15 min基本消失。条件室内两次(0 min、15 min)足底注射高渗盐水配对特定环境训练30 min,大鼠能够形成条件位置回避,单次注射或室外注射高渗盐水均不能形成条件位置回避。结论:影响痛情绪形成的因素有时间因素、线索认知以及匹配时疼痛时相,这三种因素分别指向关于痛情绪形成的两种机制:记忆机制和学习机制。
Objective: Pain-related negative emotion was an important component of pain experience.Conditioned place avoidance(CPA) apparatus was used in pain-emotion research.In this study,we explored the possible factors that influenced the formation of CPA through changing different CPA variables.Methods: We use hypertonic saline(HS) as painful-stimuli and successfully established HS-induced CPA model in rat.Results: Intraplantar injection of HS induced spontaneous pain,peaked at 5 min,recovered at 15 min.CPA was successfully established when HS hindpaw injection inside the compartment paried with conditioned context twice(0 min,15 min) within 30 min,but not with single HS injection or injection outside the chamber.Conclusion: Our results showed that time factor,cue recognition and acute pain phase have important influences on the formation of CPA model.The three influence factors point to the mechanisms underlying pain-related emotion: learning and memory.
