采用正交设计L9(34)对茶树良种湘妃翠cDNA-SRAP体系中的四因素在三个水平上进行优化试验,在25μL反应体系中,最优组合为Mg2+浓度3.0mmol/L、Taq DNA聚合酶0.1U/μL、引物浓度0.8umol/L、dNTPs浓度0.2mmol/L。以筛选出的引物组合Me6/Em6、Me7/Em6和Me7/Em7在湘妃翠、安吉白茶、龙井长叶、安茗早和湘波绿五个茶树品种材料上进行验证,表明该组合具有较好的稳定性。
By an orthogonal design L9 (3^4) the reaction system of tea tree improved variety Xiangfeicui cDNA-SRAP was optimized in terms of Mg^2+, Taq DNA polymerase, primers and dNTPs four factors and their three levels, it is optimal concluded that in a 25Q I reaction system the appropriate concentrations of Mg^2+, Taq DNA polymerase, primers and dNTPs were respectively 3.0mmol/L, 0.1U/L, 0.8u mol/L, and 0.2mmol/L. In selected primers Me6/Em6, Me7/Em6, Me7/Em7, the optimized SRAP reaction was verified with 5 tea plant varieties, Xiangfeicuh Anjibaicha, Longjingchangye, Anmingzao and Xiangbolv, showed that the composite had good stability.
