目的建立以近红外探针FA-OCC-QDs介导的裸鼠近红外体内肿瘤转移模型。方法通过化学修饰壳聚糖并包裹量子点制备具有近红外光学活性的肿瘤靶向探针FA-OCC-QDs,并结合荧光倒置显微镜、MTT法考察FA-OCC-QDs的体外肿瘤靶向性及细胞毒性,继而结合近红外成像裸鼠肿瘤转移模型、病理切片等技术研究该探针肿瘤体内靶向性。结果 FA-OCC-QDs无明显的细胞毒性(IC50=0.1368),并且具有很好的肿瘤靶向性。本研究所建FA-OCC-QDs近红外检测技术可以准确检测到体内微小的肿瘤转移灶。结论 FA-OCC-QDs可以准确诊断肿瘤转移的发生并且FA-OCC-QDs近红外成像模型的灵敏度显著高于病理切片,本研究建立了一种简单、快速、准确、实时的裸鼠活体诊断肿瘤转移的模型,该模型可能为肿瘤转移的机制研究及抗肿瘤转移药物的研发提供有利的工具。
Objective To construct NIR - imaging FA - OCC - QDs probes mediated tumors metastasis diag- nosed mouse model. Method We synthesized a biodegradable N - octyl - O, Ncarboxymethyl chitosan -folic acid conjugate ( OCC - FA) coated Quantum Dots (QDs). To examine tumor targeting efficiency and the cytotoxicity of FA - OCC - QDs, fluorescence microscope and MTY assays was used. Furthermore, NIR - imaging and pathological section were utilized to investigate the effects of OCC - FA - QDs on detecting metastasis in nude mice model of me- tastasis. Results Our result was shown that FA - OCC - QDs with tumor targeting efficiency and low cytotoxicity in vitro, and the OCC - FA - QDs mediated experimental metastatic model was successful metastatic - diagnose system for tumors micro - metastasis mechanism investigation in vivo. Conclusions We designed and synthesized a novel tumor targeting probe with NIR - imaging character and we constructed an intra - vital, precise and all - purpose an-imal model to diagnose metastatic cells in vivo. It will provide a powerful tool for basic research and drug - develop- ment of tumor metastasis.
