目的:建立免疫胶体金渗透技术检测抗子宫内膜抗体(EMAb)的新方法。方法:采用人源子宫内膜抗原,以胶体金颗粒结合的羊抗人 IgG为标记抗体,根据免疫胶体金技术原理,建立免疫胶体金渗滤技术法检测人血清抗子宫内膜抗体。分别检测73份抗子宫内膜抗体阳性血清和87份正常体检女性血清,并与酶联吸附试验( ELISA)的结果对比分析。结果:与酶联免疫吸附试验的结果对比,其符合率为90.63%,特异性为93.10%,敏感性为87.67%,交叉试验与重复试验结果显示,免疫胶体金渗透技术测定抗子宫内膜抗体具有较好的稳定性和特异性。结论:免疫胶体金渗透技术可以快速准确地测定血清抗子宫内膜抗体,便于推广普及。
Objective:To develop a new method for detection of anti endometrium antibody (EMAb) with immune colloidal gold infiltration technology. Methods:Using human endometrium antigen source, colloidal gold particles conjugated goat anti-human IgG antibody is labeled in accordance with the principles of immune colloidal gold infiltration technology, build an new method for detect endometrium antibodies in human serum. 73 positive serum anti endometrium antibodies samples and 87 serum samples of normal physical examination female serum were detected, and the result was compared with enzyme-linked assay (ELISA).Results:Compared with the result of ELISA, the consistency rate was 90.63%. The sensitivity of the assay was 93.10% and specificity was 87.67%. The preparation was stable after 6 months refrigerator storage. Conclusions:Immune colloidal gold infiltration technology can quickly and accurately measured serum anti endometrium antibody, easy to popularize.
