目的研究淋巴瘤细胞株的髓细胞白血病1(myeloid cell leukemia-1,Mcl-1)在ONO-AE-248刺激的中性粒细胞(polymorpho nuclear neutrophils,PMN)非凋亡程序化细胞死亡中的作用,为进一步确定这种新型细胞死亡模式提供线索。方法 Ficoll法新鲜分离健康志愿者外周静脉血中性粒细胞,与ONO-AE-248共同培养运用流式、RT-PCR和Western-bloting技术,对接受ONO-AE-248刺激的PMN的Mcl-1的表达进行检测。结果 ONO-AE-248刺激2 h时中性粒细胞Mcl-1 mRNA的表达明显上调,特异性抗Mcl-1单克隆抗体通过Western blotting技术检测发现其在凋亡组与非凋亡组的表达有明显差异性,表现为上升的趋势。结论 Mcl-1在ONO-AE-248诱导的非凋亡性程序化死亡中蛋白表达的上调可能是中性粒细胞凋亡和ONO-AE-248诱导的中性粒细胞非凋亡性程序化细胞死亡的关键性差异点。Mcl-1在ONO-AE-248诱导的非凋亡性程序化死亡和传统凋亡中发挥的调控作用可能是不相同的。
The aim of this study is to investigate the roles of myeloid cell leukemia-1(Mcl-1) in non-apoptotic programmed cell death(PCD) of neutrophils induced by ONO-AE-248.Polymorpho nuclear neutrophils(PMN) were separated from peripheral vein blood of health individuals,and then cocultured with ONO-AE-248.Sequentially,we detected the expression levels of Mcl-1 in ONO-AE-248-stimulated PMN by RT-PCR and flow cytometric analysis.We found that PMN could express the mRNA and protein of Mcl-1;the expression of Mcl-1 was up-regulated in neutrophils treated with ONO-AE-248.Furthermore,ONO-AE-248 could promote the non-apoptotic PCD of PMN in vitro.In conclusion,Mcl-1 may play different roles in the process of ONO-AE-248-induced neutrophil death from control and TNF-α.
