目的研究十全大补汤对肿瘤血管生成的影响,进一步探讨其抗肿瘤转移作用机制。方法采用四唑盐比色实验(MTT)、内皮细胞迁移和大鼠动脉环培养等方法检测十全大补汤对人脐静脉内皮细胞(HUVEC)的增殖、迁移和大鼠动脉环血管生成的作用。以上实验均分为空白对照组,十全大补汤0.3125,1.25,2、5,5,10mg/mL6个剂量组。结果十全大补汤各组与对照组比较,MTT的平均吸光度值升高,迁移的细胞数目减少,管腔结构形成数目减少。结论十全大补汤对内皮细胞迁移和管腔结构的形成均表现出抑制作用,这可能与其抑制肿瘤转移的作用有关。
OBJECTIVE To investigate into the effect of Shiquan Dabu Decoction on tumor angiogenesis and its mechanism of preventing tumor metastasis. METHOD MTT, endothelial cell migration and rat arterious circle cultivation were used to detect the effect of Shiquan Dabu Decoction on HUVEC hyperplasia and migration and arterious circle angiogenesis. All the experiments were made with six blank control groups: 0. 312 5, 1.25, 2.5, 5, 10 mg/mL. RESULT The average MTT absorbance increased, the number of cell migration decreased and the number of lumen structure formation decreased. CONCLUSION Shiquan Dabu Decoction has inhibiting effect on the migration of endothelial cell and lumen structure formation, which is probably related to its effect in inhibiting tumor metastasis.
