大黄鱼LcDCAF17基因的克隆与表达分析

E3泛素连接酶是细胞泛素化过程中的重要因子,DCAF17(DDB1-and CUIA—associated factor between 17 and the like protein)是E3泛素连接酶的底物受体。为了探讨DCAF17蛋白在大黄鱼性腺发育过程中的作用,从大黄鱼转录组数据库拼接出该基因,命名为LcDCAF17。LcDCAF17基因全长1733bp,开放阅读框1530bp、编码的蛋白质含509个氨基酸、分子量89.281ku、等电点5.05。LcDCAF17与其他鱼类的DCAF17同源性较高,达70%以上。荧光定量PCR分析显示LcDCAF17基因在大黄鱼稚鱼、幼鱼、成鱼的各个组织/器官中均有表达,卵巢表达量最高,精巢次之。LcDCAF17在性腺发育过程中表现出逐渐升高的趋势,在成熟期表达量最高,排空期降低。表明LcDCAF17在大黄鱼性腺发育过程中可能发挥着重要的作用。

E3 ubiquitin ligase, as a key factor in ubiquitin process, plays an important role in cell cycle regulation, and DDBl-and CUL4-associated factor between 17 and the like protein (DCAF17) is a substrate receptor protein of E3 ubiquitin ligase. To investigate the function of DCAF17 in gonad development of large yellow croaker Larimichthys crocea, DCAF17 gene ( designated as LcDCAF17 ) was identified from a comprehensive transcriptome. The full-length cDNA of LcDCAF17 is 1733 bp, containing an open reading frame (ORF) of 1530 bp which encoded a deduced protein of 509 amino acids. The molecular weight of the deduced protein was 89. 281 kDa and the isoelectric point was 5.05. The phylogenic tree showed that LcD- CAF17 shared high identities with DCAF17 from other fishes. The qPCR demonstrated that LcDCAF17 expressed in all tested tissues of Juvenile, young and adult fish and the highest expression level was in ovaries and the secondly high level in testes. With the gonad development, LcDCAF17 expression exhibited an increasing trend and peaked in maturation stage and ebbed in void phase. These results suggested that LcD- CAF17 might be involved in gonad development in L. crocea.
