为了分析不同花生四烯酸产量深黄被孢霉的差异表达基因与花生四烯酸含量的相关性,利用前期构建的不同花生四烯酸产量的深黄被孢霉消减cDNA文库,并采用反向斑点杂交技术,对该文库进行差异表达基因的鉴定,同时对鉴定的差异表达基因进行分析。结果获得了诱变前后深黄被孢霉的差异表达基因共55个。对55个阳性克隆进行测序和序列分析,结果显示25个基因在深黄被孢霉诱变前后差异表达,其中22个基因是霉菌或其他真菌基因的已知序列,其他3个基因无明显的相似序列。这些差异表达基因的鉴定可能与花生四烯酸的产生密切相关,这为进一步探讨深黄被孢霉生产花生四烯酸的分子机理研究提供一定的参考。
