The plasmids extracted from blue colonies in blue/white colonies after cloning are used to construct T vector, then the differentially displayed fragment of wheat is cloned into this T vector. Result shows that the cloning efficiency of this self-made T vector reach more than 90%. Moreover, this T vector is also practical for its convenience and low spending.
