目的考察阿魏菇多糖(pleurotus ferulae polysaccharide,PFP)对四氯化碳(CCl4)致小鼠急性肝损伤的保护作用。方法制备CCl4小鼠急性肝损伤模型,采用阿魏菇多糖(125、250和500 mg·kg-1)灌胃给药,利用试剂盒检测小鼠血清丙氨酸转氨酶(alanine aminotransferase,ALT)和天冬氨酸转氨酶(aspartate aminotransferase,AST)活性,检测肝组织中丙二醛(malondialdehyde,MDA)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)和超氧化物歧化酶(superoxide dismutase,SOD)的活力。HE染色观察肝组织病理学变化。结果阿魏菇多糖能够降低模型小鼠血清AST和ALT含量,增强肝组织SOD和GSH活力,降低肝组织MDA水平,显著改善肝组织结构,减少肝细胞坏死。结论阿魏菇多糖对CCl4致小鼠急性肝损伤有一定的保护作用。
Objective To investigate the hepato protective effects of pleurotus ferulae polysaccharide(PFP)on carbon tetrachloride(CCl_4)-induced liver injury in mice.Methods The model of acute liver injury was induced by CCl_4 in mice.The serum level of alanine aminotransferase(ALT),aspartate aminotransferase(AST) was determined respectively.Liver tissue sections were examined under a light microscope.The activity of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and content of malondialdehyde(MDA) in liver were also detected.Results Compared with control group,PFP effectively reduced the tissue lesion of liver.PFP treatment significantly decreased the serum levels of ALT,AST and hepatic level of MDA.Meanwhile,the activity of SOD and GSH-Px was increased.In addition,PFP also decreased the concentrations of TNF-α induced by CCl_4.Conclusions PFP has a significant protective effect against CCl_4-induced liver injury.
