目的 探讨刀豆球蛋白A刺激人外周血T淋巴细胞增殖的最佳浓度。方法 以人外周血制备T淋巴细胞,与刀豆球蛋白A共同培养后,采用CCK-8法检测T淋巴细胞的增殖情况。结果 不同浓度的刀豆球蛋白A对人外周血T淋巴细胞的刺激效果不同,以20μg/m L时刺激增殖效果最好。大于20μg/m L时,刀豆球蛋白A表现出对人外周血T淋巴细胞的抑制作用。结论刀豆球蛋白A对人外周血T淋巴细胞的增殖具有双重作用,且在20μg/m L时刺激增殖效果最好。
Objective To investigate the optimum concentration of concanavalin A in stimulating the proliferation of T lymphocytes of human peripheral blood. Methods T Lymphocytes were prepared from human peripheral blood, and the proliferation of T lymphocytes was determined by CCK-8 after jointly culturing with concanavalin A(Con A). Results The stimulative effect depended on the concentration of concanavalin A, and 20 μg/m L Con A had best efficacious for the proliferation of human peripheral blood T lymphocytes.When the concentration of concanavalin A 〉20 μg/m L, the proliferation of human peripheral blood T lymphocytes was inhibitted.Conclusions Concanavalin A plays a dual role in the proliferation of human peripheral blood T lymphocytes, and 20 μg/m L Con A has best proliferative effect.
