目的:探讨当归多糖对大鼠骨性关节炎(OA)滑膜和软骨形态学改变的影响。方法:4%木瓜蛋白酶关节腔注射建立大鼠OA模型,1周后,56只大鼠分成正常对照组、当归多糖对照组、模型对照组、小剂量组、中剂量组、大剂量组、阳性对照组。分组后即开始给予不同浓度的当归多糖关节腔注射,每3d1次,每次双膝各注射0.1ml,正常对照组和模型对照组注射等量生理盐水,阳性对照组给予硫酸氨基葡萄糖100mg/kg灌胃,每天1次。5周后处死动物,取关节滑膜行组织学观察并评分,切取关节软骨行大体及组织学观察,HE染色和番红O染色,并行Mankin's评分;用透射电镜观察软骨细胞和亚细胞形态变化。结果:中、大剂量组较模型对照组Mankin's评分降低(均为P〈0.01),滑膜评分降低(均为P〈0.01);较阳性对照组Mankin's评分增高(均为P〈0.01),滑膜评分无明显差别(P〉0.05)。小剂量组也有相同的作用,但较中、大剂量组作用弱(P〈0.01)。透射电镜显示小、中、大剂量组均能抑制OA软骨细胞粗面内质网和高尔基体改变,且中、大剂量组作用较小剂量组明显。结论:当归多糖关节腔注射治疗大鼠OA可促进退变软骨修复,降低滑膜炎症和纤维化程度。
Objective:To explore the effects of angelica sinensis polysaccharides(ASP)on cartilage of osteoarthritis in rats.Methods:Fifty-six rats were divided into seven groups with eight in each as follow:normal control group,0.50g/L ASP control group,model control group,0.10,0.25, 0.50g/L ASP group,and positive control group.Animal models of osteoarthritis were established by intra-articular injection of 4% papain in the knees of the rats in the latter five groups.The animals in APS group began receiving intra-articular ASP injection of different concentrations every three days.The normal and model groups received same amount of saline instead of APS.The positive control groups received 100mg/kg glucosamine sulfate by gavage everyday.Five weeks latter,all animals were sacrificed for histological examination on synovium and articular cartilage.The transmission electron microscopy were also adopted to observe the changes in articular cartilage.Results:In 0.25and 0.50g/L ASP group,Mankin's scores and synovial membrane scores significantly decreased as compared with those in model control group(both P0.01), but Mankin's scores were higher(P0.01)and synovial membrane scores had no difference(P0.05)versus that respectively in positive control group.The 0.10g/L ASP showed similar effects to 0.25and 0.50g/L ASP,but those effects were weaker(all P0.01).The transmission electron microscopy showed ASP could decrease the damage of cartilage.The 0.50g/L ASP control group showed no difference with nomal control group in every index(all P0.05).Conclusion:Intra-articular injection of ASP promotes the repairing process of degenerated cartilage of osteoarthritis in the rat models.
