高原鼢鼠(Myospalax baileyi)是一种世居青藏高原东部的地下小型穴居啮齿类动物,其生活环境最大的特点是低氧和高二氧化碳。低氧诱导因子-2α(Hypoxia Induced Factor,HIF-2α)是调节动物低氧适应的主要转录因子之一。在高原鼢鼠的HIF-2α氨基酸序列中,其他脊椎动物保守的第480位苏氨酸变异为丝氨酸。通过真核表达载体构建、定点突变和瞬时转染,真核表达获得GFP融合蛋白,为进一步研究该物种中特有的480位丝氨酸变异对HIF-2α磷酸化修饰和转录活性的影响奠定基础。
The plateau zokor(Myospalax baileyi)is one of the blind subterranean mole rats that distributed extensively in the eastern areas of the Qinghai-Tibet Plateau, living in a hypoxic-hypercapnic microenvironment. HIF-2α plays a critical role in transcriptional regulation of targeted genes during hypoxia. The serine-480(Ser480)of HIF-2α in plateau zokor is a unique amino acid variation in vertebrate. The GFP-HIF-2α fusion proteins were expressed in 293 cell through the construction of expression vector,site-directed mutagenesis and transient transfection. This work will lay the foundations for the net study of phosphorylation and transcriptional activity in plateau zokor.
