目的探讨益肾活血胶囊对动脉粥样硬化免疫炎症因子的调节作用。方法apoE—KO小鼠给予高脂饮食喂养。小鼠20周龄时随机分为模型组、益肾活血胶囊小剂量组(小剂量组)和益肾活血胶囊大剂量组(大剂量组),小剂量组给予益肾活血胶囊500mg/(kg·d),大剂量组给予益肾活血胶囊2500mg/(kg·d),模型组给予生理盐水0.2mL/d,于给药12周末处死各组试验鼠。应用ELISA法测定血清氧化低密度脂蛋白(oxLDL)含量,免疫组化检测主动脉白细胞分化抗原40(CD40);基质金属蛋白酶9(MMP9)的蛋白表达,荧光实时定量RT—PCR检测CD40、MMP9的mRNA表达情况。结果小剂量组及大剂量组oxLDL明显低于模型组(P〈0.01或P〈0.001),CD40、MMP9的表达降低,大剂量组与小剂量组相比差异有统计学意义(P〈0.05)。结论益肾活血胶囊可下调动脉粥样硬化小鼠oxLDL、CD40及MMP9的水平,对动脉粥样硬化具有一定免疫调节作用。
Objective To investigate the effect of Yishenhuoxue capsule on the expression of immune inflammatory factors in atherosclerotic mice. Methods Apolipoprotein E - knockout (apoE - KO) mice were fed with high fat diet. At 20 weeks of age, mice were divided into three groups:Model group given 0. 2 mL normal sodium daily, Yishenhuoxue capsule low dose group (low dose group) and Yishenhuoxue capsule high dose group (high dose group). Yishenhuoxue capsule were orally administered daily at 500 mg/kg and 2 500 mg/kg respectively in low dose and high dose group. After administered for 12 weeks,mice were sacrificed. Serum oxidized low density lipoprotein(oxLDL) were examined by ELISA. The expression of cell differentiation antigen 40 (CD40) and matrix metalloproteinase9 (MMP9) in aortas were examined by immunohistochemistry and real -time RT -PCR. Results The levels of serum oxLDL in Yishenhuoxue capsule groups were lower than that in model group(P〈0.01 or P〈0. 001). The expression of CD40 and MMP9 of aortas were decreased in Yishenhuoxue capsule groups. The levels in high dose group were lower than that in low dose group(P〈0.05). Conclusion Yishenhuoxue capsule could prevent atherosclerosis by lowering serum oxLDL,down- regulating the expression of CD40 and MMP - 9 in atherosclerotic mice.
