为研究鸡脾转移因子对蛋雏鸡肠道细胞因子分泌的影响,试验选取200只SPF白来航蛋鸡,随机均分成4组。各组从5日龄开始分别灌服0.1 mg/只·d、0.25 mg/只·d及1 mg/只·d剂量的鸡脾转移因子(灌服1周,停喂1周);对照组灌服生理盐水。40日龄屠宰称重,取十二指肠、空肠、回肠及直肠组织,ELISA方法检测TNF-α、IL-10的表达水平。结果表明,1 mg剂量组鸡的体重显著高于对照组(P〈0.05)。0.25 mg和1.0 mg剂量组十二指肠内TNF-α水平显著降低(P〈0.05);0.25 mg/d剂量组十二指肠、直肠的IL-10含量显著提高(P〈0.05);1 mg剂量鸡脾转移因子显著提高十二指肠IL-10含量(P〈0.05),极显著提高空肠、回肠和直肠的IL-10含量(P〈0.01)。综上结果表明,鸡脾转移因子可提高鸡肠道抗炎因子表达,抑制促炎因子分泌,改善肠道黏膜免疫屏障功能。
In order to study the effect of chicken spleen transfer factor on the expression of intestinal cytokines in laying hens and the regulation of intestinal mucosal immune function,200 SPF white leghorns were selected and evenly divided into 4 groups randomly.The groups were fed with 0.1mg/d,0.25mg/d and 1mg/d doses of chicken spleen transfer factor from 5 days old(fed a week and stop for a week).The control group was administered with physiological saline.When chicken grew to the 40 days old,duodenum,jejunum,ileum and rectum were collected and TNF-α(Tumor necrosis factor-α),and IL-10(Interleukin-10) were analyzed.The results showed that the weight of 1mg-TF-fed chicken was significantly higher than that of control group(P〈0.05).Treatment with 0.25 mg/d and 1.0 mg/d TF significantly decreased TNF-α in duodenum compared to control group(P〈0.05).IL-10 level was significantly enhanced in duodenum and rectum of chicken with 0.25 mg-TF-fed(P〈0.05),and in duodenum(P〈0.05),jejunum,ileum and rectum(P〈0.01) of chicken with1.0 mg-TF-fed.Data showed that chicken spleen transfer factors could improve the expression of anti-inflammatory factors,inhibit pro-inflammatory cytokine secretion,and improve intestinal mucosal immune barrier function.
