目的研究电针结合经颅磁刺激(rTMS)对急性脑缺血大鼠神经干细胞激活、增殖以及学习、记忆能力的影响。方法将120只雄性Wistar大鼠随机分成正常组、模型组、电针组、rTMS组和电针加rTMS组。复制急性大脑中动脉缺血模型,选取1日作为开始治疗的时间点,分别施以电针、rTMS和电针加rTMS方法处理,大鼠在各相应时间点处死前12h内每4小时腹腔注射溴脱氧尿嘧啶核苷(BrdU)1次,分别于治疗第7、14、28日进行BrdU免疫组化检测,观察鼠脑梗死面积以及鼠脑BrdU阳性细胞数,并在治疗前后对大鼠行电跳台评测。结果电针组、rTMS组和电针加rTMS组梗死侧海马齿状回颗粒细胞层(SGZ)、室管膜下层(SVZ)周围BrdU在第7、14日表达较模型组增强(P〈0.05),尤其以电针加rTMS组明显,第28日各组差异无统计学意义,电针组、rTMS组和电针加rTMS组第7、14、28日电跳台实验评分均较模型组改善(P〈0.05),尤以电针加rTMS组明显。结论电针结合rTMS治疗能促进大鼠神经干细胞的增殖,改善大鼠学习记忆能力。
Objective: To study the effect of electroacupuncture(EA) combined with transcrania lmagnetic stimulation (rTMS) on neural stem cell, learning result and memory result in rats with cerebral ischemia. Methods: One hundred and twenty male Wistar rats were randomly divided into five groups: normal group, model group, EA group, rTMS group and EA plus rTMS group. After the establishment of acute middle cerebral artery occlusion(MCAO), the rats in various groups were accordingly treated with EA, rTMS or EA plus rTMS respectively. BrdU intraperitoneal injection were given before each corresponding execute time. Then the histological sectionthe, expression of BrdU, step - down avoidance tests were observed. Results: There were a small quantity of BrdU labelled cells in SVZ and SGZ of rats in normal group. In EA, rTMS, EA Plus rTMS group, the numbers of BrdU labelled cells in SVZ, SGZ increased at 7d, 14d compared with the the model group . The expression of BrdU around SVZ and SGZ of the three treatment groups increased, espcially in the EA plus rTMS group. At 7d, 14d, 28d there was improvement in the score of step - down avoidance test in the CDE group compared with the B group, it was better in the score of step - down avoidance test in the E group than that in C and D group. Conclusion: EA plus RTMS can promote the proliferation of NSC, improve learning and memory result after cerebral ischemia.
