以间苯二酚和氰基乙酸为主要原料,经Pechmann反应,水解,甲醚化,Williamson反应合成了7种新的4-甲氧基-7-乙氧基香豆素及其衍生物。考察了影响Williamson反应的因素,Williamson反应的优化条件为,DMF溶剂中,n(4-甲氧基-7-羟基香豆素):n(卤代烃):n(碳酸钾):n(TEBA)=1:1.5:2.5:0.15,60℃反应25h,产率在57%~95%。中间产物及目标产物结构经IR,MS,^1H NMR表征得以证实。
Seven new 4-methoxy-7-ethoxycoumarin and its derivatives were synthesized from resorcinol and cyanoacetic acid by Pechmann reaction, hydrolyzation, methyl etherization and Williamson reaction. The optimum conditions for Williamson reaction were determined. When n(4-methoxy-7-hydroxycoumarin) : n (alkylhalides) : n(anhydrous potassium carbonate) : n(TEBA)=1: 1.5 : 2.5 : 0.15 and reacting at 60℃ for 25 h in DMF, the yields were between 57% - 95%. The structures of the intermediates and the products were confirmed by IR, MS and ^1H NMR.
