目的研究全反式维甲酸(ATRA)对胃腺癌细胞系SGC-7901凋亡的影响,并初步探讨其可能的分子机制。方法 ATRA处理SGC-7901细胞,Hoechst染色检测ATRA对SGC-7901细胞凋亡的影响,Western blot法检测ATRA对凋亡相关蛋白表达情况的影响。结果 ATRA可诱导SGC-7901细胞的凋亡,且下调凋亡抑制蛋白Bcl-2的表达,对促凋亡蛋白Bax的表达和酶原形式的Caspase-3的表达没有影响。结论 ATRA促进SGC-7901细胞的凋亡,其分子机制可能是下调Bcl-2的表达,使Bcl-2/Bax的值降低进而促进细胞凋亡。
Objective To study the effect of all retinoic acid ( ATRA) on the apoptosis of human gastric cancer lines SGC-7901 cells,and to investigate the possible mechanism of apoptosis. Methods SGC-7901 gastric cancer cells were treated by ATRA. Hoechst staining was used to detect the effect of ATRA on the apoptosis of SGC-7901 cells,and the effect of ATPR on the expression of apoptosis related protein was detected by Western blot. Results All trans retinoic acid ( ATRA) could induce the apoptosis of SGC-7901 cells and downregulate the expression of Bcl-2, but had no effect on the expression of pro-apoptotic protein Bax and Caspase-3. Conclusion ATRA pro-motes the apoptosis of SGC-7901 cells in gastric cancer cells. The molecular mechanism may be related to reduce the expression of Bcl-2, so that decrease the value of Bcl-2 /Bax and then promote cell apoptosis.
