PCR技术检测28个形态发育和致病性相关T-DNA插入突变体,结果所有突变体均含磷酸转移酶基因序列。观察这些突变体的生物学性状,发现15个颜色异常,8个菌落生长缓慢,2个分生孢子形态异常,2个附着胞形态异常,3个不能形成附着胞。致病性测定结果表明,9个突变体完全丧失对抗瘟(C101)和感瘟(日本晴)水稻苗的致病能力。用标准菌株1528和P131测定突变体有性世代的形成能力,结果发现3个突变体(Y34-0211、Y34-1469和Y34-0635)完全丧失产生有性世代的能力。
A total of 28 mutants from Magnaporthe grisea generated by T-DNA insertion with Agrobacterium tumifaciens-mediated transformation (ATMT) were tested with PCR for their insertional sequence. The results showed that all the genomes of these mu-tants contained the sequence of hph gene. Fifteen abnormal colony color mutants, 8 growth rate decline mutants, 2 abnormal conidiurn morphology and sporulation mutants, 2 abnormal appressorium mutants and 3 appressorium formation defective mutants were screened by observation on these mutants for their morphology and development. Nine pathogenicity defective mutants were discovered by inoculation of the conidia ffom these mutants on the susceptible rice variety (Nipponbare) and the resistant one (C101,as control). And 3 mutants (Y34-0211,Y34-1469 and Y34-0635) completely lost their ability to turn their anamorph teleomorph phase were also found by cross cultivation of these mutants with the mating type testers (P 131 and 1528).
