目的研究氯硝柳胺对肺癌细胞株顺铂敏感性的影响。方法氯硝柳胺和(或)顺铂处理人肺癌细胞株A549及顺铂耐药细胞株A549/DDP,用CCK-8法、流式细胞术检测细胞增殖、凋亡变化。结果氯硝柳胺呈剂量依赖性抑制A549、A549/DDP细胞增殖,24 h IC50分别为(2.607±0.06)、(1.135±0.09)μmol/L。氯硝柳胺联合顺铂明显增加A549/DDP细胞增殖抑制率、凋亡率(P〈0.05)。结论氯硝柳胺可抑制肺癌细胞生长和增加顺铂耐药细胞的敏感性。
Objective To investigate the effect of niclosamide on cisplatin sensitivity in lung cancer cells. Methods Human lung cancer cell line A549 and cisplatin-resistant cell line A549/DDP were treated with niclosamide and/or cisplatin, and their proliferation and apoptosis of were detected by CCK-8 assay and flow cytometry, respectively. Results Niclosamide suppressed the proliferation of A549 and A549/DDP cells in a dose-dependent manner, and IC50 at 24 h was (2.607± 0.06) μmol/L for A549 cells and (1.135 ±0.09) gmol/L for A549/DDP cells. Combined usage of niclosamide and cisplatin increased the proliferative inhibition and apoptosis of A549/DDP cells (P〈0.05). Conclusion Niclosamide can inhibit the growth of lung cancer cells and reinforce the sensitivity of cisplatin-resistant cells.
